Some suggestions:

1. if a file >2 GB gets added to an image, it should automatically switch to UDF DVD (disable ISO9660) or at least ask for that instead of burning an invalid DVD.(ISO9660 with files > 2GB)

2. It would be nice if the Filesystem properties for a new image right after startup of UltraISO could be set in the preferences of the program. For example: I prefer burning only with ISO9660 & UDF (and UDF only for DVD >2GB). So it would be nice if the user can preset the filesystem settings..

Together with suggestion 1, this would work perfectly!
(It is just like that in CopyToDVD, but I would like to abandon this program to have just one CD-burning&editing-app on my machine  :)  )

3. Just a question: what's the use of the DVD Disc checkbox in the burning dialog window?

As ISO9660 volume can hold files up to 4GB properly, UltraISO asks for switching to UDF DVD only if a file > 4GB is added.

You can do it this way:
1) Choose 'File'->'Properties' from main menu
2) Check ONLY UDF
3) Click 'Set as default' button

Then UltraISO will work as you wanted.

This is an option for NeroAPI, the next release of UltraISO will hide this option if it is not needed.

ad 1. well, at least Ultraburn created an invalid DVD if I burn files bigger than 2 GB to a DVD on-the-fly. Windows cannot read this disc. I read in a lot of posts that Nero breakes this limitation of 2GB, but still it is not part of the Standard to burn >2GB big files to a DVD. So I guess somekind of automatic disable of ISO9660 and switch to UDF only or at least a warning would be nice!

ad 2. What a shame, didn't realize it is already there.

Another question: Burning on-the fly and selecting Track-at-once will create a multi-session CD? If no, is there a way to do so with UltraiSO?

This is a bug of UltraISO when on-the-fly burning UDF volumes, it works if you save to an ISO first then burn. 4GB is not the problem.
PS: Please re-download UltraISO  from http://www.ezbsystems.com/dl.php?file=uiso8_pe.exe to avoid this problem.

No. UltraISO always finalize the disc for both DAO or TAO writing mode. So multi-session is not possible yet.
